Our job database has been working fine on a remote drive, but lately 24x7 scheduler has been unable to save to this .dat file.

Other schedulers are able to read this dat, and save. Uninstall & reinstall of 24x7 still has it unable to read the dat file.

I finally moved it to a new file on the local drive, but now after a couiple days of operation I get this message.

I have eliminated permissions (gave full) locked accounts / password (can conenct sucessfully via another computer)

Even creating a NEW dat file via another scheduler, this box is unable to read THAT file.

Any ideas?

Could it be that the file is locked by some process, perhaps backup? Sysinternals has free FileMon utility that can show all open file handles/locks. Check which processes hold a lock on the .DAT file. By default, FileMon will output will be huge, but you can set a filter by file and get nice one page view.
